Melbourne gun Christian Petracca has been cleared of serious injury but teammate Harrison Petty is among a host of AFL stars set to spend time on the sidelines.
Petty will miss four to six weeks because a foot injury, while Richmond have ruled out midfield recruit Jacob Hopper for the "medium term" with a calf strain.
Sydney have lost co-captain Callum Mills to a moderate grade calf strain and key forward Logan McDonald with a "significant medial ankle sprain".
Mills is expected to miss four to six weeks and McDonald about eight weeks.
Norm Smith medallist Petracca rolled his ankle in a tackle from Hawthorn's James Sicily late in the final quarter of the Demons' big win on Saturday.
He was sent for scans after experiencing swelling but is expected to take on Port Adelaide in the fourth-versus-second clash on Friday night.
"The scans yesterday thankfully confirmed that he just tweaked that ankle area," Melbourne club doctor Laura Lallenec said.
"He will need to get through training tomorrow and through the rest of the week to play.
"But we're fairly hopeful at this stage that there shouldn't be any issue with him playing this Friday."
Petty has impressed in his new role in attack over recent weeks but his injury will now open the door for either Ben Brown or Tom McDonald to return against Port Adelaide.
Sydney's double-blow with Mills and McDonald is another hit to a side already nursing a host of injuries to key players.
"Callum is obviously on of our on-field generals and Logan has been building into some good form, so we are disappointed to lose them for what looks like an extended period," Swans football boss Charlie Gardiner said.
"We know they will both throw themselves into their rehabilitation and do everything they can to get back as quickly as possible."
Hopper's injury is a big hit to Richmond as they chase a third straight win in Saturday night's 'Dreamtime' clash with Essendon at the MCG.
But Tigers co-captain Toby Nankervis (ankle) and young defender Tylar Young (leg) could return against the Bombers.
North Melbourne hard-man Cameron Zurhaar underwent jaw surgery at the weekend and faces a fight to prove his fitness for Saturday's encounter with Sydney.
The Kangaroos could have to make six forced changes after Jack Mahony (shoulder), Flynn Perez (concussion), Luke Davies-Uniacke and Darcy Tucker (both hamstring) were all injured in last week's loss to Port Adelaide.
Defender Aidan Corr will miss the Swans clash through suspension.
West Coast expect key forward Jack Darling to miss about a month with a fractured arm, while Jamaine Jones could play against Hawthorn on Sunday despite a broken nose.
